The mutualistic association between plant roots and arbuscular mycorrhizal (AM) fungi enhances plant nutrient acquisition (mainly phosphate) and increases plant resistance to pathogens and abiotic stresses. AM development involves cell-autonomous transcriptional reprogramming and subcellular remolding. However, this process is not synchronized; all stages of AM development are simultaneously present in the root. To unravel spatio-temporal translation during AM development, we applied tissue-specific Translating Ribosome Affinity Purification (TRAP) for Lotus japonicus roots colonized by the AM fungus Rhizophagus irregularis and combined it with RNA-seq. We identified hundreds of differentially enriched epidermis- or cortex-specific transcripts in ribosomes, including mRNAs of genes involved in AM formation. In addition, we found the mRNAs of the calcium- and phospholipid-binding Annexin1, 2, 4, and 6 strongly associated with plant ribosomes in the presence of AM fungi. The phenotypic analysis of LORE1 insertion mutants suggests a diversity of promotive or inhibitory role of these Annexins in AM development. Transactivation assays in Nicotiana benthamiana leaves indicate that different sets of transcription factor complexes activate the expression of Ann1, and Ann2, 4, and 6
