Extracellular alkalization is a hallmark of plant pattern-triggered immunity (PTI) responses. However, the mechanisms by which plant cells induce extracellular alkalization upon elicitor perception and how this process influences PTI activation remain unclear. Here, we demonstrate that the inhibition of autoinhibited H+-ATPases (AHAs) is required for elicitor-induced net H+ influx and subsequent extracellular alkalization. This alkalization is essential for immune activation mediated by diverse Pattern Recognition Receptors (PRRs) by modulating ligand-receptor interactions. Notably, PRRs transduce elicitor-triggered signaling through BOTRYTIS-INDUCED KINASE 1 (BIK1), which inhibits AHA activity by interfering with AHA-GENERAL REGULATORY FACTOR (GRF) interactions. Interestingly, this pathway is also activated in response to cell wall damage (CWD) and is essential for CWD responses. Our findings reveal a common pathway regulating extracellular alkalization to coordinate plant immune signaling.
