Effector triggered immunity (ETI) activates long-range signalling to establish broad spectrum systemic acquired resistance (SAR) to pests and pathogens with diverse lifestyles. The underlying signalling mechanisms are complex and remain poorly understood, e.g. we recently showed increased P6C (Δ1-piperideine-6-carboxylic acid) derived from oxidation of pipecolic acid depletes B6 vitamers, leading to compromised SAR. Here we report a novel luciferase reporter that captures the spatial-temporal dynamics of SAR signal generation, propagation and establishment in systemic responding leaves. JISS1 (JASMONATE INDUCED SYSTEMIC SIGNAL 1), encodes an ER localised protein of unknown function. JISS1:LUC reporter dynamics are unaffected in classical SAR mutants, but both jasmonate biosynthesis and perception mutants abolished JISS1:LUC signalling and SAR to Pseudomonas syringae. Furthermore, ETI elicited jasmonate-dependent systemic surface electrical potentials which required both glutamate receptors and JISS1. Neither JISS1 loss-of-function nor glutamate receptors mutants altered SAR to Pseudomonas syringae. Thus, jasmonate signalling, usually associated with antagonism of defence against biotrophs, is crucial to the initiation and establishment of SAR to P. syringae and activation of systemic surface electrical potentials
